بهبود حافظه فضایی موش های صحرایی نر مدل پارکینسونی پس از تیمار با سلول های بنیادی چربی و عصاره برگ رزماری

سابقه و هدف: باتوجه به نقش نوروپروتکتیو عصاره رزماری، در مطالعه حاضر درمان همزمان پیوند سلول­های بنیادی بافت چربی و عصاره بر اختلال حافظه موش ­های پارکینسونی مورد بررسی قرار گرفت.
مـواد و روش ­ها: موش ­های نر مدل پارکینسونی با تزریق دوطرفه 6-OHDA آماده شدند. به جسم سیاه گروه شم نرمال سالین تزریق شد. گروه عصاره+محیط کشت، گاواژ عصاره رزماری را 14 روز قبل از آسیب تا 8 هفته پس از آن دریافت کردند و به دنبال آن تزریق داخل وریدی محیط کشت انجام شد. گروه عصاره+سلول با عصاره گاواژ شدند و سپس سلول تزریق شد. آموزش ماز آبی موریس، یک هفته قبل و بعد از آسیب و همچنین آزمون به ­خاطرآوری در هفته­ های 4 و 8 پس از آسیب انجام شد.
نتایج: مسافت طی شده و تأخیر زمانی یافتن سکو در روزهای آموزش قبل از آسیب تفاوت معنی­ داری بین گروه ­ها نشان نداد. یک هفته پس از بروز آسیب توانایی یادگیری در حیوانات گروه آسیب نسبت به شم کاهش معنی­ داری داشت (0/05<P). نتایج تست­ های به ­خاطرآوری هفته ­های 4 و 8 مشابه یکدیگر بودند. مدت زمان پیدا کردن سکو و مدت سپری شده در ربع هدف در موش ­های آسیب دیده به ­ترتیب افزایش و کاهش معنی­ داری نسبت به شم نشان داد (0/05<P). گروه­ های عصاره+محیط کشت و عصاره+سلول، به ­ترتیب کاهش و افزایش معنی­ داری را در مدت زمان پیداکردن سکو و مدت زمان سپری شده در ربع هدف نسبت به گروه آسیب نشان دادند (0/05<P).
نتیـجه ­گیری: سلول­درمانی به همراه تجویز خوراکی عصاره رزماری باعث بهبود اختلال حافظه در بیماری پارکینسون می­ گردد.
