The Evaluation of BDNF Expression and Spatial Memory after VPA Administration in Animal Model of Hippocampal Degeneration

Background: Hippocampal degeneration which causes cognitive deficits and decrease of neurotrophins. Valproic acid (VPA) as a histone deacetylase inhibitor could protect nervous system from neuronal degeneration with neurotrophin modulations. The present study sought to investigate the effect of VPA on hippocampal level of BDNF, histopathological analysis of CA1 area of hippocampus and evaluation of spatial memory in animal model of TMT induced hippocampal degeneration.
Materials and Methods: In order to induce hippicampal degeneration, TMT (8mg/kg) was injected intra-peritoneally (i.p) to the rats. The three test groups (TMT+VPA) received 100, 200 and 400 mg/kg of VPA respectively and the TMT+Saline group received normal saline for 14 days after TMT intoxication. The Morris water maze (MWM) test was conducted in order to investigate the spatial memory. Then, the hippocampal level of BDNF was measured using ELISA and histopahological evaluation of hippocampus was done.
Results: VPA was decreased distance and latency time to hidden platform in learning blocks and increased spent time in target quadrant in probe test after TMT intoxication. In addition, the hippocampal level of BDNF and amelioration of cell damages in CA1 area of hippocampus were increased in the VPA-treated groups compared with TMT+Saline group.
Conclusion: VPA having sufficient neuroprotective effects could be ameliorated TMT induced cognitive deficits in rats.
