Study of some neurotrophin genes expression in human adipose stem cells cultured in serum-containing and serum-free media

Background: Since, the FBS is immunogenic for human and may transfer infection in the case of transplantation.  In this study, proliferation and survival rate, neurotrophin genes expression of cultured human adipose stem cells (hASCs) in serum-free and serum containing media were compared. The results were prepared the way to design an appropriate substitute for FBS used in traditional culture.
Materials and Methods: In this experimental study,subcutaneous adipose tissue was extracted from abdomen of 15 cesarean women. After mechanical and enzymatic digestion, stem cells were cultured in α-MEM containing10% of fetal bovine serum and serum-free medium. At fourth passage, the stemness related properties of cells were determined by flow cytometry and also their differentiation into adipocyte and osteocyte. Cell proliferation and survival were done by using hemocytometry and MTT [3- (4,5-Dimethyltiazol-2-yl) -2,5-Diphenyltetrazolium bromide] methods respectively. The expression of neurotrophic factor  genes of hASCs were examined by RT-PCR.
Results: The cells had a positive response to CD44, CD73, CD90, CD105 markers and negative response to CD34 and CD45 markers and had the ability to differentiate into adipocyte and osteocyte. Results of cell count and MTT showed that survival and proliferation of cells cultured in serum-based medium for 48 hours were significantly increased compared to cultured cells in serum free medium. In addition, serum-based culture caused a significant increase of BDNF and NT-3 expression as compared to cultured cells in serum-free medium.
Conclusions: The results can be matched to find a replacement for FBS, with the same trophic effect and no harms. More suitable cells can be provided for transplantation with serum deletion and optimize culture medium.
