تغییرات بیان پروتئین گیرنده گلوتاماتی NMDA و کانال Pannexin-1 در بافت آنتروم معده موش‌های صحرائی مبتلا به دیابت نوع 2 و تحت درمان با انسولین

زمینه و هدف: شواهد نشان می­ دهد گیرنده ­های گلوتاماتی در بروز آسیب ­های گوارشی ناشی از دیابت نقش دارند. از این رو، در این مطالعه به الگوی بیان پروتئین گیرنده گلوتاماتی N-متیل-D-آسپارتات (NMDA) و کانال pannexin-1(Panx-1)، که ارتباط عملکردی مهمی با گیرنده‌های گلوتاماتی دارد، در معده موش­ های دیابتی نوع 2 پرداختیم. همچنین تأثیر درمان طولانی مدت با انسولین را بر تغییرات احتمالی پروتئین­ های مذکور بررسی کردیم.
روش‌ها: در این مطالعه تجربی 18 سر موش صحرایی نر ویستار با وزن تقریبی 90-80 گرم به طور تصادفی به 3 گروه (هر کدام 6 موش) تقسیم شدند: 1) کنترل با رژیم غذایی استاندارد، 2) دیابت نوع 2: تحت رژیم غذایی پرچرب به مدت 3 ماه + تک دوز استرپتوزوتوسین ( 35 میلی‌گرم بر کیلوگرم)، و 3) دیابت نوع 2 + درمان با انسولین. محتوای پروتئینی گیرنده NMDA و همچنین Panx-1 با استفاده از روش وسترن بلات شناسایی و بین گروه ها مقایسه شد.
یافته‌ها: در معده موش­های دیابتی نوع 2 بیان پروتئین Panx-1 به طور معنی ­دار کاهش داشت (0/001P<)، در حالی که بیان گیرنده NMDA (0/001P<) افزایش معنی­داری را نشان داد. درمان با انسولین سطح بیان پروتئین Panx-1 را در موش‌های دیابتی بهبود بخشید، علاوه بر این، درمان با انسولین باعث کاهش سطح بیان گیرنده NMDA در موش‌های دیابتی شد (0/05P<).
نتیجه‌گیری: نتایج حاضر حاکی از تغییرات مهم سطوح پروتئینی گیرنده NMDA و کانال Panx-1 و نقش احتمالی آنها در بروز آسیب­ های حسی-حرکتی و ترشحی مرتبط با دیابت است. اصلاح تغییرات فوق ممکن است در بهبود آسیب­ های عملکردی ناشی از دیابت مزمن نوع 2 نقش داشته باشد.
