Role of sacular endothelium on dilatory responses to Isoproterenol in respiratory arterial rings

History and Objectives: Due to discrepancies on the role of endothelium on dilation of vascular beta-adenoceptor and possible involvement of L-arginine/nitric oxide, the present experimental study was undertaken on the main pulmonary arteries and its first branches in rat.

Materials and Methods: An experimental study on 6 wistar rats (250-300 g) was carried out. By applying a heavy blow, the animals where anesthetized, heads were cut off with surgical knife and heart and lung were separated. In cold Krebs solution the main artery and the first pulmonary branches were separated from the attached tissues. Arteries were cut in to 3-4 mm sections, placed on a hook in a tissue bath and at 37°C and one gram force was applied to stretch the tissue. Isoproterenol (ISO) in concentration of 100pmole and 10 micromole in the presence and absence of N-omega-nitro-L-arginine methylester (L-NAME, Nitric Oxide inhibitor) were applied to arterial rings with and without pulmonary endothelium.

Results: Isoproterenol causes dilation of pulmonary arterial rings (Main and branches) in the presence and absence of endothelium. Arteries were constricted with 100 nonmole phenylephrine. Maximum dilatory response to 10 ?mole of isoproterenol in the arterial rings was 93.04%±1.25 and in the first branches were 77.21%±2.80 (Unpaired test, P<0.001). The main pulmonary arterial ring response was higher than its first branches. L-NAME at low ISO concentrations reduced the dilatory response of pulmonary ring in the presence of intact endothelium, however it did not have an effect at higher concentration of ISO.

Conclusion: The results shows that dilatory response of rat pulmonary arteries to ISO does not require the presence of NO. However presence of endothelium may facilitate the dilatory response of ISO by releasing basal NO and subsequently elevating basal cGMP levels in vascular smooth muscles. Further research is needed to measure cGMP levels in order to determine the released NO concentrations.
