اثر تجویز حادّ دکسترومتورفان، کوئینیدین و ترکیب دکسترومتورفان/ کوئینیدین بر آستانه تشنجات کلونیک و تونیک ناشی از پنتیلن تترازول (PTZ) در موش سوری

سابقه و هدف: دکسترومتورفان به­ عنوان یک ضدّ­سرفه غیراپیوئیدی، اثرات حفاظت عصبی دارد. ترکیب دکسترومتورفان با کوئینیدین، متابولیزه­شدن سریع دکسترومتورفان به دکستروفان را کاهش می ­دهد. هدف از مطالعه حاضر بررسی اثر تجویز حادّ دکسترومتورفان، کوئینیدین و ترکیب دکسترومتورفان/ کوئینیدین بر آستانه تشنجات کلونیک و تونیک ناشی از پنتیلن تترازول (PTZ) در موش سوری می‌باشد.
مواد و روش ­ها: در مجموع، 84 سر از موش ­های سوری نر نژاد NMRI (25-20 گرم، 7 سر موش در هر گروه) در این مطالعه استفاده شدند. غلظت­ های مختلف دکسترومتورفان (5، 10، 25 و 50 میلی ­گرم بر کیلوگرم)، کوئینیدین (10، 20 و 30 میلی­ گرم بر کیلوگرم) و ترکیب دکسترومتورفان/ کوئینیدین (20/5، 20/10، 20/25 و 20/50 میلی­ گرم بر کیلوگرم) 30 دقیقه قبل از القای تشنج به ­صورت داخل‌صفاقی تجویز شدند. از انفوزیون داخل‌وریدی PTZ برای القای تشنج استفاده شد و مدت زمان وقوع کلونوس عمومی و کشیدگی تونیک اندام عقبی، ثبت و به دوز آستانه تشنج تبدیل شد.
نتایج: غلظت 30 میلی­ گرم بر کیلوگرم کوئینیدین آستانه تشنج تونیک را به ­صورت معنی­ داری افزایش داد (0/05<P). غلظت­ های 25 و 50 میلی­ گرم بر کیلوگرم دکسترومتورفان آستانه تشنج کلونیک (0/05<P) و تونیک (0/001<P) را به صورت معنی­ داری افزایش دادند. غلظت 50/20 دکسترومتورفان/ کوئینیدین آستانه تشنج کلونیک و تونیک را در مقایسه با گروه دکسترومتورفان کاهش داد (0/001<P).  
نتیجه­ گیری: نتایج نشان داد که اثر متفاوت دکسترومتورفان بر آستانه تشنج کلونیک و تونیک ممکن است به ­خاطر تفاوت در حساسیت مغز قدامی و مغز خلفی نسبت به دکسترومتورفان باشد. همچنین کاهش اثر دکسترومتورفان در حضور کوئینیدین ممکن است به ­خاطر تغییر در متابولیسم دکسترومتورفان باشد.
