بررسی اثر لیتیوم کلراید بر بهبود اختلال حافظه کاری و فضایی در مدل حیوانی تشنج القاء شده با پنتیلن تترازول

سابقه و هدف: حملات تشنجی مکرر منجر به آسیب نورونی گسترده و بروز اختلالات شناختی مانند زوال حافظه و یادگیری می­ شود. تجویز مکرر یا تک دوز پنتیلن تترازول با القاء تشنج در موش ­های صحرایی همراه است. این مطالعه به بررسی اثر حمایت کننده عصبی لیتیوم کلراید بر نقص حافظه کاری و فضایی ناشی از پنتیلن تترازول می­ پردازد.

مواد و روش­ ها: جهت القاء کیندلینگ ناشی از پنتیلن تترازول، دوز تکرار شونده (۴۰ میلی­ گرم/کیلوگرم وزن بدن حیوان) به­مدت ۵ روز و به ­صورت درون صفاقی تزریق شد. پس از مشاهده ۵ مرحله ­تشنج، گروه­های PTZ+Li۲۰، PTZ+Li۴۰ و PTZ+Li۸۰ به­ ترتیب دوزهای ۲۰، ۴۰ و ۸۰ میلی­ گرم/کیلوگرم لیتیوم کلراید و گروه PTZ+Saline محلول نرمال سالین را به­مدت ۱۴ روز دریافت کردند. آزمون­ های ماز آبی موریس و ماز Y جهت بررسی حافظه ­کاری و حافظه فضایی انجام شد.

نتایج: در مقایسه با گروه کنترل، گروه PTZ+Saline کاهش رفتارهای تناوبی طی ماز Y و افزایش مدت زمان و مسافت پیمایش تا رسیدن به سکوی مخفی طی ماز آبی موریس را نشان داد. از طرفی، حیوانات دریافت کننده لیتیوم کلراید، به­ ویژه در دوز حداقل، درصد بالاتری از رفتارهای تناوبی در ماز Y، به ­همراه کاهش مدت زمان و مسافت پیمایش مسیر را در ماز آبی در مقایسه با گروه PTZ+Saline نشان دادند.

نتیجه­ گیری: اثرات محافظت کنندگی عصبی لیتیوم سبب بهبود حافظه کاری و حافظه فضایی در مدل حیوانی تشنج القاء شده با پنتیلن تترازول می­ گردد.
