مقایسه پاسخهای رفتاری مدلهای درد نوروپاتیک در موش صحرایی نربالغ

<p align="justify"><strong>سابقه و هدف:</strong> آسیب عصبی یکی از مهمترین عوامل ایجادکننده دردهای نوروپاتیک می باشد که با علایمی مثل درد خودبخودی، آلودینیا و هایپرآلژزیا همراه است. با توجه بر اینکه مکانسیمهای زمینه ساز دردهای نوروپاتیک کاملاً شناخته شده نیستند، تحقیقات وسیعی در این خصوص در حال انجام است. لذا این مطالعه به منظور مقایسه پاسخهای رفتاری مدل های درد نوروپاتی CCI و SNI با گروه شاهد انجام گرفت. </p><p align="justify"><strong>مواد و رو ش ها:</strong> این تحقیق به روش تجربی انجام و از موشهای صحرایی نر بالغ نژاد Dawley Sprague در محدوده وزنی 230 تا 280 گرم استفاده گردید. حیوانات بصورت تصادفی به 4 گروه 8 تایی تقسیم شدند و بر اساس وزن بدن به وسیله پنتوباربیتال سدیم با دوز mg/kg 50 و به صورت تزریق درون صفاقی بیهوش شدند. آنگاه عمل جراحی بر اساس مدلهای CCI و SNI و گروه شاهد روی حیوانات صورت گرفت. سپس پاسخهای رفتاری حیوانات نسبت به محرکهای غیردردناک حرارتی (تست حباب استن) و مکانیکی (تست Von Frey ) و محرکهای دردناک حرارتی (تست Radiant Heat ) و مکانیکی (تست Pin Prick ) طی روزهای صفر( قبل از عمل جراحی) و 3، 7، 14، 21 و 28 بعد از عمل جراحی بررسی و از آنالیز آماری ANOVA تکراری جهت مقایسه نتایج حاصل از تستهای رفتاری استفاده گردید. </p><p align="justify"><strong>یافته ها :</strong> پاسخهای رفتاری گروههای CCI و SNI در مقایسه با گروه شاهد نسبت به تحریکات مکانیکی و حرارتی غیردردناک افزایش معنی د اری داشت که نشاندهنده آلودینیای حرارتی و مکانیکی بود. همچنین حساسیت به محرکهای مکانیکی و حرارتی آسیب ر سان در گروههای CCI و SNI نسبت به گروه شاهد افزایش معنی د اری نشان داد که حاکی از هایپرآلژزیای حرارتی و مکانیکی بود. بعلاوه، حیوانات گروه SNI در مقایسه با گروه CCI آلودینیای مکانیکی را در روز سوم با دامنه بالاتری نشان دادند ( p<0.05 ). اما حیوانات گروه CCI در مقایسه با گروه SNI آلودینیای حرارتی را در روز هفتم شدیدتر نشان دادند ( P<0.05 ). </p><p align="justify"><strong>نتیجه گیر ی:</strong> حیوانات در مدلهای درد نوروپاتی SNI و CCI حساسیت شدیدی به محرکهای حرارتی و مکانیکی غیر دردناک (پدیده آلودینیا) و محرکهای حرارتی و مکانیکی دردناک (پدیده هایپرآلژزیا) نشان دادند، بطوریکه پدیده آلودینیای مکانیکی در حیوانات گروه SNI برجسته تر از حیوانات گروه CCI بود. </p>
<p align="justify"><strong>Background: </strong>Neuropathic pain syndromes are changes resulted from damage to neuronal pathways which is characterized by spontaneous burning pain with accompanying allodynia and hyperalgesia. The mechanisms underlying neuropathic pain are poorly understood. The present study explores behavioral characteristics of the neuropathic pain models chronic constriction injury (CCI) and spared nerve injury (SNI). </p><p align="justify"><strong>Materials and Methods: </strong>Experiments were performed on four groups (n= 8) of male Sprague-Dawley rats (230-280 g). Anesthesia was initially induced with sodium pentobarbital (i.p.) at a dose of 50 mg/kg. The CCI model was made by loose ligation of the sciatic nerve. Also a lesion of two of three terminal branches of the sciatic nerve leads to a SNI model. The animals were tested for behavioral responses cold-and mechano-allodynia and heat-and mechano-hyperalgesia. The cold and mechanical stimulations in the cold- and mechano-allodynia phenomena were applied through acetone and von Frey filament respectively. Pin-prick and radiant heat were applied as thermal and mechanical stimulations in the heat- and mechano-hyperalgesia respectively. Behavioral tests were conducted on the animals prior to surgery (the day 0), and 3, 7, 14, 21 and 28 days post-operation. </p><p align="justify"><strong>Results: </strong>Our results indicate that, in comparison to the controlled group, the rats in both SNI and CCI groups reveal an obvious difference in behavioral responses. Although the SNI, compared to the CCI group were more sensitive to mechano-allodynia shortly after surgery (p<0.5) however, both groups share a similar pattern of behavior. In the heat-hyperalgesia testing, again, the animals in the CCI and SNI groups behaved differently than those in the controlled group, but no variation was evident among the test groups, themselves. </p><p align="justify"><strong>Conclusion </strong>: These findings clearly show that the two neuropathic models produce abnormal pain-related disorders in the rats. A major feature of the SNI model was the very marked hypersensitivity to normally innocuous mechanical stimuli. </p>
