پروتئین کیناز C مهار بازسازی میلین در دمیلیناسیون موضعی القا شده در کیاسمای بینایی موش را میانجی گری می کند

<p><strong>سابقه و هدف:</strong> پروتئین های مهاری موجود در میلین با فعال کردن PKC، ترمیم آکسونی در مغز بالغین را محدود می کنند.اگر چه مهار PKC، توانایی پروتئینهای میلین را در مهار رشد آکسونی کاهش می دهد، ولی نقش و مکانیسم آن در مهار بازسازی میلین تاکنون ناشناخته است. با توجه به درگیر شدن دستگاه بینایی در بیماری MS، هدف از این مطالعه بررسی اثر مهار PKC در بازسازی میلین کیاسمای موش های دمیلینه شده با لیزولیستین می باشد. </p><p><strong>مواد و روش ها:</strong> با تزریق یک میکرولیتر لیزولستین دمیلیناسیون در کیاسمای موش نر القاء شد و تزریق روزانه داخل بطنی مهار کننده پروتئین کیناز (C (GÖ6976 تا 14 روز پس از القاء ادامه یافت. الگوی دمیلیناسیون و رمیلیناسیون در کیاسمای بینایی با رنگ آمیزی اختصاصی میلین و با استفاده از ثبت پتانسیل برانگیخته بینایی (VEP) انجام شد.</p><p><strong>نتایج:</strong> در حیوانات دریافت کننده لیزولسیتین به تنهایی بیشترین دمیلیناسیون در روزهای 3 و 7 پس از تزریق مشاهده شد و رمیلیناسیون کمی هم در روز 14 دیده شد، اما در حیواناتی که با مهارگر پروتئین کینازC نیز تیمار شده بودند، دمیلیناسیون کمی در روز 3 پس از تزریق مشاهده شد و رمیلیناسیون در روزهای 7 و 14 افزایش یافت. در داده های حاصل از ثبت VEP نیز در گروه آسیب، بیشترین تاخیر موج p در روز های 3 و 7 بود که در روز 14 برگشت به سمت نرمال داشت، ولی در گروه تیمار میزان تاخیر موج p در روزهای 3 و 7 به شکل قابل ملاحظه ای کاهش داشت. </p><p><strong>نتیجه گیری:</strong> به نظر می رسد فعالیت پروتئین کیناز C در مهار ترمیم درون زاد میلین نقش بارزی داشته و مهار آن به عنوان یک راه کار درمانی جدید برای پیشبرد فرایند رمیلیناسیون در بیماری هایی مثل ام.اس. قابل استفاده باشد. </p>
<p><strong>Background:</strong> Axon regeneration in adult CNS is limited by the presence of inhibitory proteins associated with myelin. Although blocking PKC activity attenuates the ability of CNS myelin to inhibit neurite outgrowth, the role as well as mechanisms underlying the remyelination inhibition in CNS are still largely unknown. Considering the role of PKC in axonal regeneration and the vulnerability of optic chiasm in multiple sclerosis (MS), we assessed the effect of PKC inhibition on remyelination of lysolecithin induced demyelinated optic chiasm.</p><p><strong>Materials and Methods:</strong> Demyelination was induced by stereotaxic intra-chiasmatic injection of 1µl lysolecithin (%1) in male mice. Intracerebroventricular daily injection of a PKC inhibitor (GO6976) was done for 14 days post-lesion. Demyelination and remyelination patterns in optic chiasm were confirmed through histological verification and electrophysiological study using Luxol fast blue staining and visual evoked potentials (VEP) recording, respectively.</p><p><strong>Results:</strong> In lysolecithin treated animals, demyelination was mostly marked at days 3 and 7 post-lesion and an incomplete remyelination occurred at day 14 post-lesion. VEP recording showed increased P-latency at the days 3 and 7 post-lesion while it partially decreased at day 14. Following the inhibition of PKC, while the extent of demyelination and P-latency slightly decreased at the days 3 and 7 post-lesion, it recovered at day 14. VEP recording data were confirmed by histological verification.</p><p> <strong>Conclusion:</strong> Inhibition of PKC activity could represent a potential therapeutic approach for stimulating the remyelination process in the context of multiple sclerosis. </p>
