ارزیابی اثرات محافظت کنندگی در برابر انجماد سرم جنین گاو و ترهالوز بر میزان زنده‌مانی سلول های بنیادی اسپرماتوگونی بز

سابقه و هدف: سلول­ های بنیادی اسپرماتوگونی (SSCs: Spermatogonial Stem Cells)، اساس اسپرماتوژنز هستند؛ زیرا ظرفیت خودنوسازی و تمایز به اسپرماتوزوآ را دارند. انجماد، متداول­ترین روش حفظ طولانی‌مدت SSCs است. هدف از مطالعه حاضر، بررسی اثرات محافظت­ کنندگی در برابر انجماد سرم جنین گاو (FBS: Fetal Bovine Serum) و ترهالوز بر SSCs بز بود.
مواد و روش­ ها: SSCs از بیضه بز نابالغ توسط هضم آنزیمی و حذف تمایزی جداسازی شدند. سلول­ ها به 9 گروه تقسیم شدند. گروه کنترل شامل SSCs بدون عوامل محافظت­ کننده انجماد بود. در گروه­های تیمار 1، 2، 3 و 4؛ غلظت 10 درصد FBS و غلظت ­های مختلف ترهالوز (0، 50، 100 و 200 میلی­ مول) و در گروه ­های تیمار 5، 6، 7 و 8؛ غلظت 20 درصد FBS و غلظت­ های مختلف ترهالوز (0، 50، 100 و 200 میلی ­مول) به ­ترتیب استفاده شد. میزان زنده‌مانی سلول ­ها بلافاصله پس از جداسازی، افزودن مواد محافظ انجماد و یخ­ گشایی ارزیابی شد. ماهیت سلول‌ها توسط رنگ‌آمیزی ایمنوسیتوشیمی علیه آنتی‌ژن PGP9.5 تأیید شد. داده‌ها توسط آزمون آنالیز واریانس یک­ طرفه تحلیل گردیدند.
نتایج: میزان زنده‌مانی SSCs در تیمارهای مختلف، متعاقب افزودن FBS و ترهالوز بلافاصله پس از جداسازی، مشابه سلول ­های زنده بود. علاوه‌بر این، بیشترین میزان زنده‌مانی SSCs پس از یخ ­گشایی در محیط انجماد حاوی 10 درصد FBS و 200 میلی­مول ترهالوز مشاهده شد (0/05>P).
نتیجه­ گیری: نتایج نشان داد که انجماد در FBS 10 درصد توأم با 200 میلی­مول ترهالوز به ­عنوان روش مؤثر برای حفاظت از انجماد SSCs بز عمل می ­کند.
