بررسی شیوع مایکوپلاسما ژنیتالیوم و مایکوپلاسما هومینیس در بین زنان مبتلا به عفونت واژینال در شهر رباط کریم طی سال 1392

سابقه و هدف: مایکوپلاسما ژنیتالیوم و هومینیس از عوامل مهم واژینوز محسوب می­ شوند و شناسایی آنها توسط روش سنتی کشت دادن با مشکل مواجه است. هدف از انجام این تحقیق، بررسی شیوع این باکتری­ ها با به کارگیری تکنیک PCR در مقایسه با کشت در زنان مبتلا به عفونت می ­باشد.

مواد و روش ­ها: برای انجام این مطالعه مقطعی 250 نفر بیمار مراجعه ­کننده به بیمارستان ­های امام خمینی و امام زمان(عج) استان تهران و 150 نفر فرد سالم مورد بررسی قرار گرفتند. DNA استخراج شده از نمونه ­ها به­ عنوان الگو جهت تکثیر ژن کد کننده 16s rRNA با استفاده از پرایمرهای اختصاصی در دو واکنش PCR جداگانه استفاده شد.

نتایج: نتایج به ­دست آمده نشان داد که کشت مایکوپلاسما ژنیتالیوم و مایکوپلاسما هومینیس به ­ترتیب در 38 و 46/8 درصد موارد مثبت بود، درحالی­ که با مطالعه نمونه­ ها به روش PCR به­ ترتیب 62 و 77/2 درصد از نمونه ­ها مثبت گزارش شدند. نتایج نشان­ دهنده آن است که برای شناسایی مولکولی این باکتری­ ها PCR از دقت، حساسیت و اختصاصیت ویژه­ای برخوردار است، به­ طوری­ که نمونه­ های کشت منفی نیز با این روش قابل تشخیص می ­باشند.

نتیجه ­گیری: برای شناسایی عوامل عفونی در مبتلایان به واژینوز، روش مولکولی PCR به­ طور معنی­ داری نسبت به­ کشت از حساسیت بالاتری برخوردار بوده و با به ­کارگیری پرایمرهای اختصاصی می ­توان این عوامل بیماری ­زا را در حد جنس و گونه با دقت بیشتری مورد شناسایی قرار داد.
