fa بررسی تأثیر تمرین تناوبی شدید و مصرف کورکومین بر بیان ژن PGC-1αو ERR-α میتوکندریایی کاردیومیوسیت‌ موش‌های نر مدل سکته قلبی Investigation the effect of high intensity interval training (HIIT) with curcumin supplementation on mitochondrial cardiomyocyte PGC-1α and ERR-α gene expression in male stroke model rats عمومى General پژوهشي Research <div style="text-align: justify;">سابقه و هدف: آنفارکتوس میوکارد یک وضعیت نکروزی حادّ عضله قلب است. بنابراین هدف از پژوهش حاضر، بررسی تأثیر تمرین تناوبی شدید و مصرف کورکومین بر بیان ژن PGC-1α و ERR-α میتوکندریایی کاردیومیوسیت‌ موش‌های نر مدل سکته قلبی بود. مواد و روش‌ها: در مطالعه تجربی حاضر، 32 سر موش صحرایی نر مبتلا به آنفارکتوس میوکارد به‌صورت تصادفی به چهار گروه تمرین تناوبی با شدت بالا، مکمل، کنترل و تمرین به همراه مکمل تقسیم شدند. تمرین در دو گروه شدت بالا و تمرین به همراه مکمل شامل اجرای هشت هفته فعالیت 4دقیقه‌ای با شدت 90-85 درصد VO2max بود. کورکومین نیز به میزان mg/kg 15 استفاده شد. بیان ژن PGC-1α و ERR-α میتوکندریایی با استفاده از روش  Real-time PCRبه‌دست آمد. ﺩﺍﺩﻩ‌ﻫﺎ از طریق ﺁﺯﻣﻮن شاپیرو - ویلک و تحلیل واریانس یک‌طرفه ﺁﻧﺎﻟﻴﺰ ﺷﺪﻧﺪ. نتایج: آزمون تحلیل واریانس یک‌طرفه، تفاوت معنی‌داری در بیان ژن PGC-1α و ERR-α میتوکندریایی کاردیومیوسیت‌ موش‌های نر مدل سکته قلبی در گروه‌های پژوهش نشان داد (0/001=P). نتایج آزمون توکی بیانگر افزایش معنی‌دار بیان ژن PGC-1α و ERR-α در گروه تمرین و تمرین به همراه مکمل نسبت به دو گروه کنترل و مکمل بود (0/001=P). بین دو گروه کنترل و مکمل نیز تفاوت معنی‌داری وجود نداشت. نتیجه‌گیری: تمرین تناوبی به‌تنهایی و در تعامل با کورکومین باعث افزایش بیان ERRα و PGC-1α در بافت قلب موش‌های نر مدل سکته قلبی می‌شود. به‌نظر می‌رسد تمرین تناوبی و مصرف کورکومین می‌تواند راه مناسبی برای افزایش بیوژنز میتوکندریایی و بهبود عملکرد قلب در نظر گرفته شود.</div> <div style="text-align: justify;">Background: Myocardial infarction is an acute condition of myocardial necrosis. So, this study aimed the effect of high intensity interval training with curcumin supplementation on mitochondrial cardiomyocyte PGC-1α and ERR-α gene expression rats. Materials and Methods: 32 male rats were randomly divided into 4 groups of HIIT, supplement, control and HIIT with supplement training in the two groups of high intensity and HIIT with supplement included performing 8 weeks of 4-minute activity with an intensity of 85-90% VO2max. Curcumin was also used at 15 mg / kg. Mitochondrial PGC-1α and ERR-α gene expression was obtained by real-time PCR. Data were analyzed by ANOVA and Shapiro-Wilk at P≤0.05. Results: One-way analysis of variance test showed a significant difference in the expression of mitochondrial cardiomyocyte PGC-1α and ERR-α genes in male myocardial infarction mice in the study groups (P=0.001). The results of Tukey test showed a significant increase in PGC-1α and ERR-α gene expression in the HIIT and HIIT groups with supplementation compared to the control and supplement groups (P=0.001). There was no significant difference between the control and complement groups. Conclusion: The results of this study showed that interval training alone and in interaction with curcumin increases the expression of ERRα and PGC-1α in the heart tissue of male stroke model mice.
