fa تعیین ژنوتیپ آپولیپوپروتئین E در بیماران دیابتی با عارضه قلبی - عروقی با تکنیک PCR اختصاصی آلل Determination of apolipoprotein E genotyping in diabetic patients with cardiovascular diseases with allele specific PCR method عمومى General پژوهشي Research <div style="text-align: justify;">سابقه و هدف: آپولیپوپروتئینE  (ApoE) یکی از چند لیپوپروتئین مهم در متابولیسم لیپیدهاست که پژوهش‌های فراوانی در رابطه با آن انجام شده است. در این پژوهش‌ها به نقش این پروتئین در بیماری‌هایی مثل آلزایمر، بیماری‌های قلبی - ‌عروقی، دیابت و بیماری‌های نقص ایمنی مانند ایدز پرداخته شده است. ژن این آپولیپوپروتئین چندشکلی و دارای سه رونوشت یا آلل مختلف به نام‌های e2، e3 و e4 است. این سه آلل به‌صورت شش ژنوتیپ E2E2، E2E3، E2E4، E3E3، E3E4 و E4E4 با فراوانی‌های مختلفی در جمعیت وجود دارند. بسته به ویژگی‌های جمعیتی، جغرافیایی و نژادی، این ژنوتیپ‌ها با فراوانی متغیر در جمعیت دیده می‌شوند که در هر حال بیشترین فراوانی مربوط به E3E3 است. در این تحقیق ژنوتیپ آپولیپوپروتئین E در بیماران دیابتی با عارضه قلبی - عروقی با تکنیک PCR اختصاصی آلل تعیین شد. مواد و روش‌ها: این مطالعه روی 107 نمونه خون بیمار دیابتی دارای بیماری‌های قلبی - ‌عروقی ساکن شهر کاشان انجام گرفت. برای تعیین ژنوتیپ این ژن، از تکنیک PCR اختصاصی آلل با پنج پرایمر و سپس بررسی الگوی باندها در الکتروفورز استفاده شد. نتایج: داده‌ها نشان می‌دهد که در جمعیت موردمطالعه یعنی بیماران دیابتی دارای بیماری‌های قلبی - ‌عروقی ساکن شهر کاشان، بیشترین ژنوتیپ E3E3 است. همچنین فراوانی آلل‌های e2، e3 و e4 به ترتیب برابر با 1/5، 79 و 9/15 درصد است. نتیجه‌گیری: این مطالعه نشان می‌دهد که تأثیر متوسط این ژنوتیپ بر لیپوپروتئین‌های پلاسما یا انواع CVD وجود دارد، بنابراین می‌تواند برای پیش‌بینی و پیشگیری از بیماری‌های مرتبط با این ژنوتیپ مفید باشد.</div> <div style="text-align: justify;">Background: Apolipoprotein E (ApoE) is one of the several important lipoproteins in lipid metabolism, and many studies have been conducted on it. In various studies, the role of this protein in diseases such as Alzheimer's disease, cardiovascular disease, diabetes and immune deficiency diseases such as AIDS have been addressed. The gene is a polymorphic apolipoprotein with three transcripts or different alleles called e2, e3 and e4. These three alleles have six genotypes E2E2, E2E3, E2E4, E3E3, E3E4 and E4E4 with different frequency in the population. Depending on the demographic, geographic and racial characteristics of these genotypes, they are seen with varying frequency in the population, which is in any case the highest frequency related to E3E3. In this study determinated apolipoprotein E genotyping in diabetic patients with cardiovascular diseases with allele specific PCR method. Materials and Methods: This study was performed on 107 blood samples of diabetic patients with cardiovascular disease living in Kashan. To determine the genotype of this gene, allele specific PCR with five primers was used and then the pattern of bands in electrophoresis was used. Results: The results show that in this population (diabetic patients with cardiovascular disease living in Kashan), as in most Iranian and world populations, the most genotype is E3E3. Also, the frequency of e2, e3 and e4 alleles is 5.1, 79 and 15.9%, respectively.
