fa بررسی اثر اسانس بهارنارنج (Citrus aurantium L.) بر شاخص‌های سلامت کبدی در مدل حیوان آزمایشگاهی Investigation of the effect of essential oil from Citrus aurantium L. flowers on liver health parameters in a laboratory animal model medicine, paraclinic medicine, paraclinic پژوهشي Research <div style="text-align: justify;">سابقه و هدف: در فرهنگ غذایی ایرانی، گل‌ها و اسانس بهارنارنج در قالب مربا و عرقیجات در مقادیر بالا مصرف می‌شود. در طب سنتی ایرانی در مورد اثرات عرق بهارنارنج روی کبد نگرانی‌هایی ذکر شده است. هدف از مطالعه حاضر، بررسی اثر اسانس بهارنارنج بر شاخص‌های سلامت کبدی در مدل حیوانی می‌باشد. مواد و روش‌ها: اسانس بهارنارنج با کلونجر استخراج و توسط کروماتوگرافی گازی - جرمی آنالیز شد. سپس با دوزهای (g/kg ۲ و ۱، 0/5) به‌مدت سه روز به‌صورت تک‌دوز از طریق داخل‌صفاقی یا خوراکی برای شش گروه پنج‌تایی موش صحرایی تجویز شد. برای دو گروه کنترل (g/kg۱) روغن زیتون (حامل اسانس) تجویز شد. ۴۸ ساعت بعد از آخرین تجویز، نمونه‌گیری خون و بافت کبد جهت تعیین آلانین امینو ترانسفراز(ALT) ، اسپارات امینو ترانسفراز(AST) ، لاکتات دهیدروژناز (LDH) و گلوتاتین (GSH) انجام شد و مطالعات هیستوپاتولوژیک صورت گرفت. نتایج: تغییرات اصلی اسانس، لینالیل استات، ترپینن-۴-ال، لینالول و فنتیل الکل بود. تجویز صفاقی اسانس در دوزهای بالاتر از (g/kg ۱) موجب افزایش معنی‌دار AST، ALT و LDH شد. این اسانس در غلظت‌های بالاتر از (g/kg ۱) در تجویز خوراکی و داخل‌صفاقی باعث التهاب بافت کبدی شد. نتیجه‌گیری: بررسی آنزیم‌های AST، ALT و LDH و داده‌های هیستوپاتولوژی نشان داد که این اسانس در دوزهای بالا، دارای سمیت خفیف کبدی می‌باشد. این عوارض ناشی از استرس اکسیداتیو نبوده، احتمالاً از طریق سمیت مستقیم و غیراکسیداتیو ایجاد می‌شود.</div> <div style="text-align: justify;">Background: In Persian food culture, Citrus aurantium L. (bitter orange) flowers and essential oil are consumed as jams or plant distillate in high doses. Some concerns were mentioned in Persian traditional literature about their effects on the liver. This study aimed to investigate the effect of the essential oil from C. aurantium L. flowers on liver health parameters in a laboratory animal model. Materials and Methods: The essential oil was extracted by Clevenger-type apparatus and analyzed by gas chromatography-mass spectrometry. Six groups of rats (n=5) received intraperitoneal or oral essential oil with the doses of (0.5, 1 or 2 g/kg) for 3 days. The two control groups received olive oil (the vehicle). Forty-eight hours after the last administration, serum and liver samples were prepared and alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and glutathione (GSH) were determined. The liver histopathological changes was investigated. Results: The main ingredients of the essential oils were linalyl acetate, terpinene-4-ol, linalool and phenethyl alcohol. The intraperitoneal administration of essential oil at doses higher than (1 g/kg) caused a significant increase in AST, ALT and LDH levels. Inflammation of the liver tissue was observed at oral and intraperitoneal doses higher than (1 g/kg). Conclusion: The changes in AST, ALT and LDH levels showed that at high doses of the essential oil had a mild liver toxicity and the histopathologic data confirmed this.
