اثر والپروئیک اسید بر بیان ژن‌های مسیر خارجی (DR4, DR5, FAS, FAS-L, TRAIL) و مسیر داخلی آپوپتوز (BAX, BAK and APAF1, Bcl-2, and Bcl-xL)، میزان زنده بودن سلول و آپوپتوز سلولی در سرطان کبد رده سلولی PLC/PRF5

سابقه و هدف: استیلاسیون و دِاستیلاسیون هیستون، نقش مهمی در نسخه‌برداری و بیان ژن بازی می‌کنند. وضعیت استیلاسیون پروتئین‌های هیستونی و غیرهیستونی به‌وسیله آنزیم‌های هیستون استیل ترانس فراز (histone acetyl-transferases, HATs) و هیستون دِاستیلاز (histone deacetylases, HDACs) تعیین می‌شود. مهارکننده‌های آنزیم‌های هیستون دِاستیلاز، اثرات مولکولی و خارج سلولی مختلفی القا می‌کنند که منجر به ایفای نقش ضدسرطانی آن‌ها می‌شود. این تحقیق به منظور بررسی اثر داروی والپروئیک اسید بر بیان ژن‌های مسیر خارجی و مسیر داخلی آپوپتوز، میزان زنده بودن سلول و آپوپتوز در سرطان کبد رده سلولی PLC/PRF5 طراحی شد.
مواد و روش‌ها: سلول‌های سرطانی کبد رده PLC/PRF5 کشت داده شدند و پس از این‌که هم‌پوشانی سلول‌ها به حدود 80 درصد رسید، سلول‌ها با تریپسین جمع‌آوری گریدند و پس از شستشو، در پلیت با غلظت 105× 3 کشت داده شدند. پس از 24 ساعت، محیط کشت با محیط حاوی داروی والپروئیک اسید تیمار شدند (گروه کنترل فقط DMSO دریافت کردند). 24، 48 و 72 ساعت پس از تیمارکردن، به منظور تعیین میزان زنده بودن سلول، سلول‌های آپوپتوتیک و بیان ژن، به‌ترتیب تکنیک‌های MTT، فلوسیتومتری و Real-time انجام شد.
نتایج: والپروئیک اسید به‌طور معنی‌داری باعث مهار رشد سلولی، القای آپوپتوز، کاهش بیان ژن‌های Bcl-2, and Bcl-xL و افزایش بیان ژن‌های DR4, DR5, FAS, FAS-L, TRAIL, BAX, BAK and APAF1 گردید.
نتیجه‌گیری: به‌نظر می‌رسد والپروئیک اسید نقش آپوپتوزی خود را از طریق مسیرهای آپوپتوزی داخلی و خارجی در سرطان کبد ایفا می‌کند.
