بررسی اثرات غلظت سیتوتوکسیک تستوسترون، پروژسترون و استرادیول بر بیان ژن ضدّ متاستازی CD82/KAI1 در سلول های سرطانی گلایوبلاستومای مغزی

سابقه و هدف: مطالعات نشان می ­دهند که هورمون­ های جنسی می­ توانند بر متاستاز سلول ­های سرطانی سیستم عصبی در سطح سلولی و مولکولی تأثیر بگذارند. با این حال مکانیسم سلولی و مولکولی عملکرد استروئید­های جنسی بر متاستاز سلول های سرطانی سیستم عصبی در موارد زیادی واضح و آشکار نیست. هدف از این مطالعه بررسی اثرات غلظت سیتوتوکسیک تستوسترون، پروژسترون و استرادیول بر بیان ژن ضدّ متاستازی CD82/KAI1 در سلول­­ های گلایوبلاستومای مغزی (A172) می­ باشد.
مواد و روش­ ها: در این مطالعه تجربی- آزمایشگاهی، رده سلولی A172 از بانک سلولی انستیتو پاستور تهیه شده و به گروه­ های شاهد و در مواجهه با غلظت ­های سیتوتوکسیک هورمون­های تستوسترون، پروژسترون و استرادیول تقسیم ­بندی شدند. سطح نسبی بیان ژن CD82/KAI1 با استفاده از روش Real time PCR اندازه­گیری شد. درنهایت داده­ ها با استفاده از روش آماری آزمون واریانس یک ­طرفه مورد تجزیه و تحلیل قرار گرفت.
نتایج: سطح بیان نسبی ژن ضدّ متاستازی CD82/KAI1 در سلول­ هایA172  در گروه ­های دریافت­ کننده دوزهای توکسیک تستوسترون و پروژسترون و استرادیول نسبت به گروه کنترل دچار کاهش معنادار شد (به ­ترتیب 0/001<P، 0/05<Pو 0/001<P).
نتیجه ­گیری: تستوسترون و پروژسترون و استرادیول می­ توانند با دوز سیتوتوکسیک، سبب کاهش بیان نسبی ژن ضدّمتاستازی CD82/KAI1 شده، بدین ­ترتیب باعث افزایش متاستاز در سلول­های سرطانی مغز شوند.
